CU Integrated Biomedical Imaging Facility

Confocal Microscope

LSM 510 META confocal scanning system with three lasers:

  • Argon 458/477/488/514 nm
  • Green HeNe 543 nm
  • Red HeNe 633 nm

Zeiss AxioPlan 2IE MOT motorized upright microscope, with the following objectives:

  • Plan Neo 40x oil 1.3 n.a. DIC
  • Plan 63x 1.4 n.a. DIC
  • Achroplan 40x 0.8 n.a. water DIC for live cell work
  • C-Apochromat 40x 1.2 n.a. DIC for fluorescence correlation spectroscopy
  • Plan Apo 10x
  • Plan Neo 20x 0.75 n.a. DIC

META scanning module with eight single-channel detectors.

Coherent Chameleon ULTRA Ti:S laser (690 – 1040nm) for multi-photon excitation.

Two non-descanned detectors for multi-photon work.

Becker & Hickl spectroscopy system for fluorescence correlation spectroscopy (FCS) and time-correlated single photon counting (TCSPC).

Motorized stage for time series experiments.

We can image with most standard fluorophores, in either single-photon or multi-photon modes. The META multi-channel system enables separation of fluorophores with closely-spaced emission peaks by linear un-mixing.

We have software for FRET, FRAP and FLIM, ratiometric imaging, and complex time series experiments.

Additional equipment:

  • Two optical stretchers with IPG Photonics 10 W 1060 nm IR fiber lasers for cytomechanics studies.
  • Metamorph 3-D reconstruction on a Nikon Eclipse 800.
  • High-resolution transmitted light and epi fluorescence imaging on a Zeiss Axioskop II with 100x Plan Apochromat objective and SPOT RT camera.
  • Total Internal Reflection Fluorescence (TIRF) Imaging

Facility Usage Fees:

Assisted:                               $50.00/hr

Unassisted:                          $25.00/hr

This facility was constructed with support from C06 Grant RR17417-01 from the NCRR, NIH.

Contact Dr. Jensen-Smith to obtain training for use of the confocal microscope and for more information about our terms of use agreement. Once trained, reserve time for confocal use with our online Reservations System.
